Title: Proteomic analysis on a MALDI-MS platform

Srinivas Iyer, Bioscience Division (B-8), Los Alamos National Laboratory

Advances in mass spectrometry have facilitated the analysis of protein expression and modification changes in biological systems. Several approaches using different ionization mechanisms are available, each with a set of pros and cons. MALDI-MS shot into prominence for protein and proteomics analysis. As with all ionization methods, MALDI has its advantages and disadvantages. For a variety of reasons, my laboratory is completely dependent on this platform and we have optimized it for the type of applications we encounter most. Here we will discuss the rationale for our instrumentation choice, review possible workflows using currently available instrumentation at LANL and present examples from projects using our proteomics capability. Time permitting, we will also discuss intact protein measurements, unconventional small molecule analysis and future plans for expansion of this capability.
